Culmerciclib rechallenge for HR+ HER2- breast cancer
This trial tests a new drug called Culmerciclib for women with advanced HR-positive, HER2-negative breast cancer that has stopped responding to a CDK4/6 inhibitor with hormone therapy. The goal is to see if giving this drug again can help when other treatments no longer work.

This trial tests if adding a targeted drug (CDK4/6 inhibitor) to hormone therapy works better than hormone therapy alone for women with a certain type of breast cancer (HR+/HER2-). The study measures the activity of tumor cells (Ki-67) to help decide treatment.

This early study (Phase 1) tests RLY-2608, a targeted cancer drug for tumors with a PIK3CA mutation. It studies the drug alone and combined with hormone therapy (and sometimes CDK4/6 or CDK4 medicines) to see safety, dose, and whether it can shrink tumors.
