XELOX with Dostarlimab after chemoradiation for rectal cancer

This trial tests whether adding the immunotherapy drug dostarlimab (a checkpoint inhibitor) to standard chemotherapy (XELOX) after chemoradiation improves outcomes for patients with locally advanced rectal cancer that is not MSI-high. You may be eligible if your tumor has specific genetic features called pMMR/MSS or MSI-low, and you have not received prior chemotherapy or radiation.

Who can take part

  • You have rectal adenocarcinoma that is within about 16 cm of the anal opening
  • Your cancer stage is T3-4 N0 M0, or any N+ with at least 3 lymph nodes >0.5 cm or 1 node >1 cm on imaging
  • Your tumor is pMMR/MSS or MSI-low (not MSI-high)
  • Your performance status is 0 or 1 (fully active or slightly limited)
  • You have not had prior chemotherapy or radiation for this cancer
  • Your blood counts and kidney/liver function are within defined normal limits
