Elacestrant and exemestane for HR+/HER2- breast cancer that has spread

This trial tests combining elacestrant (a new hormone therapy) with exemestane (a standard hormone therapy) for people whose HR+/HER2- breast cancer has spread and who have already tried a CDK4/6 inhibitor with anastrozole or letrozole. It may help if your tumor shows certain activity on a special PET scan.

Who can take part

  • You are 18 or older.
  • Your breast cancer is hormone receptor-positive (HR+) and HER2-negative, and has spread or is locally advanced and cannot be removed by surgery or cured by radiation.
  • You are a good candidate for hormone therapy (no emergency symptoms from the cancer).
  • You have at least one tumor that can be measured or bone lesions that can be evaluated on imaging, and those tumors show high activity on a specific type of PET scan called FES-PET.
  • You have already tried a CDK4/6 inhibitor (like palbociclib, ribociclib, or abemaciclib) together with anastrozole or letrozole for at least 12 months, and your cancer stopped responding or progressed during or within 28 days of finishing that treatment.
  • You have not had any other treatment after that CDK4/6 inhibitor combination, and you are in generally good health (able to do daily activities).
