- Your cancer is 'microsatellite stable' (MSS)—a genetic feature your doctor can confirm from prior testing
- Your cancer has progressed or caused unacceptable side effects from first-line chemotherapy (oxaliplatin plus 5-FU or similar)
- Your blood counts, kidney function, and liver function are adequate; your heart pumps normally
- You have at least one measurable tumor on imaging and are in good enough health (ECOG 0–1: able to do light or no activity)
- You have had no chemotherapy, radiotherapy, or major surgery in the 4 weeks before starting the trial
