New drug plus rapamycin and chemo for HER2-positive breast cancer

This Phase 3 trial tests whether a new HER2-targeting drug (inetetamab) combined with rapamycin and standard chemotherapy works better than the control approach for HER2-positive metastatic breast cancer. It is mainly for people whose tumor genetics suggest a specific “PAM pathway” problem.

Who can take part

  • You are a woman and older than 18
  • Your breast cancer is HER2-positive based on test results (IHC and/or FISH)
  • Your cancer is confirmed invasive breast cancer that has come back or spread (metastatic)
  • Your tumor has a mutation in the PI3K/Akt/mTOR pathway genes (related to the PAM pathway)
  • You can do activities at about a normal level (ECOG score 0–2) and expect to live at least 6 months
  • Your heart and organ tests are good enough to start treatment (heart pumping strength LVEF at least 50% and acceptable blood counts/liver tests)
