Study drug RC48-ADC plus S-1 for advanced HER2 stomach cancers
Treatments studied
Part of Cancer, Digestive system clinical trials.
This Phase 1/2 study tests RC48-ADC (an antibody-drug treatment) together with S-1 (a chemotherapy medicine) in people with advanced stomach or esophagus/gastroesophageal junction cancer. It’s mainly for cancers with HER2 testing results of IHC 2+ and FISH-negative, and aims to find a safe dose and how well the treatment works as a first treatment option.
Summary written for real people, not researchers, by Clin2.
Who can take part
- You are 18 or older and can sign informed consent
- Your cancer is confirmed as stomach, gastroesophageal junction, or esophageal adenocarcinoma that cannot be fully removed by surgery (or has spread)
- You have not had chemotherapy for advanced disease before, or you progressed/relapsed within 6 months after finishing earlier chemotherapy
- Your tumor HER2 test is IHC 2+ and FISH-negative (based on standard lab tests)
- Your general health is good (ECOG 0–1) and doctors expect you to live at least 3 months
- If you can become pregnant (or if you can father a child), you must use reliable birth control during the study and for 12 weeks after the last dose

This trial tests a combination of three drugs (targeted therapy, immunotherapy, and chemotherapy) before and after surgery for locally advanced HER2-positive stomach cancer. It aims to see if this treatment can improve outcomes for patients whose cancer has not yet spread.
This study tests two new immunotherapy drugs (KN026 and KN046) with or without chemotherapy (XELOX) for people with a specific type of advanced stomach cancer that makes too much HER2 protein. The goal is to see if this combination can shrink tumors or slow the cancer.
This trial is testing a combination of four medicines (RC48, adebrelimab, apatinib, and S-1) as a pre-surgery treatment for people with locally advanced stomach cancer that is HER2 positive. The goal is to shrink the tumor before surgery and possibly improve outcomes.
This trial tests a combination of three drugs—RC48 (a targeted therapy), sintilimab (an immunotherapy), and S-1 (a chemotherapy pill)—as a first treatment for advanced HER2-positive stomach cancer that has not been treated before. The goal is to see if this triple therapy works better than current options.
This trial is testing a combination of two drugs (RC48 and AK104) as the first treatment for advanced HER2-positive stomach cancer. The goal is to see if this combination can shrink tumors and help people live longer.
This trial tests a new combination of drugs (RC48, sintilimab, and SOX chemotherapy) for people with stomach or gastroesophageal junction cancer that cannot be removed by surgery, has not been treated before, and has a specific HER2 score (1+ or 2+). The goal is to shrink the tumor enough to possibly allow surgical removal.
