This trial compares a new oral drug, palazestrant (OP-1250), to standard hormone therapies for people with ER+/HER2- advanced breast cancer that has worsened after prior hormone therapy. It tests whether palazestrant can better control the cancer.

Who can take part
- You must be an adult (18 or older).
- Your breast cancer must be ER-positive (ER+) and HER2-negative (HER2-), and it has spread or cannot be cured with surgery or radiation.
- You must have taken a CDK4/6 inhibitor with hormone therapy before, and you were on your last hormone therapy for at least 6 months.
- You need to be well enough to do daily activities (ECOG score 0 or 1).
- If you are a man or a woman who is premenopausal or perimenopausal, you must be willing to take a medicine to stop your ovaries from making hormones (called a GnRH agonist).
