Who can take part
- You have breast cancer that is hormone receptor-positive (HR+) and HER2-negative.
- You have a confirmed mutation in the BRCA1, BRCA2, or PALB2 gene (from a blood or tumor test).
- Your cancer is advanced (locally advanced or has spread to other parts of the body) and you have not yet received any systemic treatment for this advanced stage.
- You are generally well enough to do most daily activities (ECOG 0 or 1).
- You do not have untreated brain metastases, spinal cord compression, or certain other serious medical conditions.

This trial tests a drug called talazoparib (a PARP inhibitor) for advanced breast cancer that has a specific DNA repair defect (HRD) or a BRCA mutation. The goal is to see if the drug works better when the patient's tumor is tested for this defect just before treatment.
