- Rectal cancer confirmed by biopsy, located within 5 cm of the anus
- Early-stage cancer (T1-3, N0, M0)—tumor hasn't spread to lymph nodes or distant organs
- No previous cancer treatment
- Good overall health (ECOG performance status 0-1) and expected to live at least 3 months
- Normal or near-normal blood counts, liver, kidney, and heart function
- Willing to use contraception during treatment and for 12 months after
