Liver-targeted treatment for genetic leptin-related obesity
Part of Hormones & metabolism clinical trials.
This study is for people with a rare genetic condition that causes severe obesity and high appetite, called leptin receptor deficiency or related leptin problems. The trial tests a new approach that targets the liver to help with weight control, and it aims to see if this can improve their condition.
Summary written for real people, not researchers, by Clin2.
Who can take part
- You or your child must have a confirmed diagnosis of leptin receptor deficiency (a genetic problem that affects the hormone that controls appetite) or a related genetic cause of low leptin with lipodystrophy (abnormal body fat distribution).
- You can be on current treatments like setmelanotide or metreleptin – that's fine, or you can be untreated.
- Your obesity must not be due to a different genetic reason.
- You must not have liver disease from other causes, such as heavy alcohol use.
- Females who can become pregnant must not be pregnant or planning pregnancy during the study.

This program provides access to an experimental drug (REGN4461) for people with rare conditions related to a problem with the hormone leptin, which helps control appetite and body weight. It is an expanded access program, meaning it's a pathway for patients who have no other treatment options.
This trial tests an open-label study of metreleptin in people with partial lipodystrophy, a condition where fat is lost from some areas of the body. The goal is to see if the drug can help manage related health problems.
This trial tests a new drug called mibavademab for people with severe obesity caused by changes in the leptin (LEP) gene. It aims to see if the drug can help with weight loss in children, teens, and adults who have this rare genetic condition.
This trial tests whether an experimental drug (LY3437943) can help people with obesity or overweight lose weight. It compares the drug to a placebo (inactive shot), and you must be willing to give yourself daily injections.
This study uses multiple types of health data (“multi-omics”) to help choose a personalized treatment plan for people with obesity-related fatty liver disease. It may help match treatment to your biology to improve outcomes.
This clinical trial tests new treatments for adults who are overweight or have obesity. The goal is to help people lose weight and improve their health.
