This Phase 1/2 trial tests a combination of an EZH2 inhibitor (tulmimetostat) with a PD-1 blocker (pembrolizumab) for advanced non-small cell lung cancer that has stopped responding to prior PD-1 treatment. It may help shrink or control cancer and is mainly aimed at people who have limited treatment options left.

Who can take part

  • Be 18 years or older and sign consent (including permission for future research).
  • Have advanced non-small cell lung cancer confirmed by biopsy/pathology, and measurable tumors on scans.
  • Have had prior treatment with chemo (platinum-based) and/or immunotherapy, and then your cancer must have progressed on an anti–PD-1/PD-L1 medicine (like pembrolizumab) after at least 2 doses.
  • Have had at least one tumor tissue sample (archived sample or a new biopsy) for study testing.
  • Have basic blood and organ test results that meet specific cutoffs (including neutrophils, platelets, hemoglobin, kidney, liver, and blood clotting), and you should be able to do daily activities (ECOG 0–1).
