Advanced Solid Tumors With MAPK Pathway Mutations

Who can take part

  • You have an advanced solid tumor (spread or locally advanced) that is getting worse.
  • Tumor testing from the past 5 years shows a MAPK pathway gene mutation (Part 1) or a KRAS G12C mutation (Part 2).
  • For non-small cell lung cancer (Part 2), your tumor must NOT have certain other gene changes like EGFR, ALK/ROS1/RET, NTRK, BRAF V600E, or MET exon 14 skipping.
  • You’ve tried standard treatments—Part 1 means standard treatment is no longer working, not tolerated, or not available; Part 2 requires at least one prior systemic treatment (including a KRAS G12C drug).
  • You must have “measurable disease” on scans (Part 2), plus provide tumor tissue (archival or optional fresh biopsy) and blood for genetic testing.
  • Your health status is high enough to participate (ECOG 0–1) and your organs are functioning adequately; you must also follow birth control/reproductive rules.
