This trial tests different sequences of antibody-drug conjugates (a type of targeted chemotherapy) for people with ER-positive, HER2-low or ultra-low metastatic breast cancer that has stopped responding to hormone therapy and has progressed on Enhertu. You must have had 1–4 prior chemotherapies for metastatic disease.

Who can take part
- You must have breast cancer that is estrogen or progesterone receptor positive (ER+ or PR+) and is HER2-low or ultra-low.
- Your cancer must have grown despite hormone therapy, and you must have taken a CDK4/6 inhibitor (like Ibrance) before.
- You must have received between 1 and 4 prior chemotherapy treatments for metastatic disease. Prior antibody-drug conjugate (ADC) therapy and PARP inhibitor therapy each count as one line.
- You must have received Enhertu (T-DXd) before and stopped it either because it stopped working or caused intolerable side effects.
- You must have measurable disease by a CT or MRI, or bone-only disease with a lytic (destroyed) lesion. Blastic (dense) bone lesions alone are not allowed.
- You must be 18 or older, have adequate organ and blood cell counts, and if applicable, use effective birth control.
