- Age 18–75 years old
- Locally advanced rectal cancer (stage II or III) confirmed by biopsy, with the tumor within 10 cm of the anus
- No distant spread (metastasis) of cancer
- Good overall health (ECOG performance status 0–1: able to carry out normal activities)
- Adequate blood counts, kidney, and liver function
- No prior chemotherapy, radiation to the pelvis, or immunotherapy for this cancer
