This Phase 2 trial tests whether adding different medicines—either avelumab or hydroxychloroquine, with or without palbociclib—can eliminate dormant “leftover” breast cancer cells. It’s meant for people with hormone-receptor–positive breast cancer who have no visible cancer now, but still have detectable dormant cells in the bone marrow.

Who can take part
- You must have had all standard breast cancer treatments completed, and your only remaining treatment allowed during the study is hormone therapy (endocrine therapy).
- Your bone marrow test after finishing treatment must show detectable dormant tumor cells, confirmed by the study’s central lab.
- Your breast cancer must be ER-positive and HER2-negative (and not ER-negative with PR-positive only).
- You must currently be taking adjuvant hormone therapy, and you must be within 2–7 years of starting it. If randomized to hydroxychloroquine, tamoxifen usually needs to be switched to an aromatase inhibitor at least 21 days before starting study treatment.
- If you previously took a CDK4/6 inhibitor (a targeted hormone-related drug), you must have stopped it at least 6 months before screening.
- You must be able to swallow and absorb hydroxychloroquine well, and you must not have certain heart rhythm problems, uncontrolled illness, severe lung disease, or eye problems from retinopathy.
