Who can take part
- You have rectal adenocarcinoma confirmed by biopsy, and your tumor testing must show MSS and “MGMT silenced” (no MGMT protein and MGMT gene turned off).
- Your cancer must be locally advanced but still removable with curative surgery (possibly including a more extensive pelvic surgery).
- Your body is strong enough for treatment: performance status 0–1 (up and active most of the day) and you must have blood counts good enough for chemo.
- Your scans must show no spread to other organs (no metastatic disease) and specific imaging features must match the trial’s “locally advanced” definitions.
- You must be able to provide stored tumor tissue for central lab testing.
- You must not have certain exclusions, including prior pelvic radiation and a body metabolism problem called DPD deficiency.
