Who can take part
- You have HR-positive and HER2-negative breast cancer confirmed by a lab
- Your cancer test shows a PI3K-pathway change (PIK3CA mutation, PTEN loss, or AKT1 mutation)
- Your cancer is advanced and not suitable for cure (locally advanced or metastatic) and you can show progression after prior treatments, or you progressed while/on CDK4/6 plus hormone therapy
- You have measurable cancer on scans (or only certain bone-only spots are allowed for safety groups)
- You can take pills by mouth and you can fast for clinic visits (over 8 hours) on fasting days
- Your health tests are adequate (for example, blood counts, liver/kidney tests) and your performance status is 0–1
