This Phase 1/2 study tests how well a new treatment combination (VS-6766, abemaciclib, and fulvestrant) works for metastatic or hard-to-remove breast cancer that is hormone-driven (ER/PR positive) and HER2 negative. It is mainly designed to find the safest dose and then see whether it can control the cancer after it has progressed on certain prior medicines.

Who can take part

  • You have breast cancer that tests positive for hormone receptors (ER/PR) and tests negative for HER2.
  • Your cancer has spread (metastatic) or come back locally in a way that can’t be removed with surgery.
  • Your disease has grown or progressed after treatment that included a CDK4/6 medicine, and you have also progressed on fulvestrant.
  • You can take medicines by mouth, and you have no major uncontrolled health problems; basic blood and organ tests must be acceptable.
  • You are able to use effective birth control during the study and for a period after treatment ends (pregnancy is not allowed).
  • You have not had certain recent treatments (chemo/immunotherapy within 21 days, endocrine/biologic drugs within 14 days, and radiation or major surgery within 14 days).
