Pioglitazone plus metformin for high-risk mouth lesions

This trial studies whether a two-medicine diabetes drug combination can reduce or control risky early changes in certain mouth and throat lesions. You may be asked for small tissue samples (biopsies) before and after 12 weeks to see how the lesions respond.

Who can take part

  • You’re 18 or older, and you can give written consent.
  • A biopsy has already shown abnormal cell growth (hyperplasia or dysplasia) in a high-risk area of the mouth or throat, such as the tongue, floor of mouth, or oropharynx.
  • You have measurable oral white or red patches (leukoplakia and/or erythroplakia) that can be measured in two directions, and the main spot must be big enough for a small 4 mm punch biopsy.
  • You can take a twice-daily tablet regimen for 12 weeks (swallowing tablets whole).
  • You have no major medical “organ function” problems based on recent blood tests done within 14 days of starting the study.
  • If you can become pregnant, you agree to reliable birth control during the study.
