Who can take part
- Age 18–80 with good overall health (PS score 0–1, meaning you can do light activities)
- Confirmed colorectal cancer with KRAS G12C mutation (a specific genetic change) and certain other genetic markers (NRAS and BRAF wild-type, MMR/MSS status)
- Locally advanced cancer that hasn't been treated with systemic (whole-body) therapy before; if you have metastases (cancer spread), they must be removable with surgery and you haven't received targeted therapy for them
- Adequate organ function: blood cell counts, liver and kidney function, heart function (LVEF ≥50%), and normal blood clotting within acceptable ranges
- No active infections, uncontrolled high blood pressure, heart disease, bleeding disorders, recent blood clots, or other serious illnesses that would make treatment unsafe
- No prior targeted therapy for this cancer (including other G12C inhibitors, bevacizumab, or cetuximab)
