This trial tests a personalized DNA vaccine made from your tumor’s DNA (neoantigens) to help your immune system fight the cancer. It is designed for children and young adults with certain recurrent or newly diagnosed brain/spine tumors, after they have had other standard treatments and tumor tissue is available for testing.

Who can take part

  • Aged 12–25 and have a qualifying brain or spine tumor (high-grade glioma or diffuse midline glioma newly diagnosed, or other pediatric CNS tumors with recurrent or resistant disease).
  • You must have tumor tissue available for sequencing to find target “neoantigens” (tumor markers that can be recognized by the immune system).
  • Your doctor expects you may live more than 24 weeks for the tissue-testing step and more than 12 weeks for the treatment step.
  • You must be able to consent (or have a guardian who can), and agree to use effective birth control during the study.
  • You can’t have certain serious vaccine reactions, uncontrolled medical problems, or active immune-suppressing conditions that would block an immune response.
  • If the vaccine cannot be made (no usable neoantigen found), you cannot continue into treatment.
