This trial studies whether adding palbociclib to cetuximab works better than cetuximab by itself for head and neck cancer that is growing again after an anti–PD-1/PD-L1 treatment. It is for people whose tumor has a specific gene change (CDKN2A loss) and who have not had too many prior treatments.

Who can take part

  • You have recurrent or metastatic head and neck squamous cell cancer that is HPV-unrelated.
  • Your tumor test shows a CDKN2A loss-of-function change (like a mutation or loss of both copies).
  • Your cancer has measurable spots on scans (meeting minimum size rules).
  • Your cancer grew despite treatment with an anti–PD-1/PD-L1 drug (alone or with other drugs).
  • You have had 0 to 3 prior treatment “lines” for recurrent/metastatic disease.
  • You are able to use effective birth control during the study (and for a set time after) and you meet basic blood/organ health requirements.
