Lenvatinib plus letrozole versus fulvestrant for metastatic breast cancer

This Phase 2 study compares a combination medicine (lenvatinib plus letrozole) against fulvestrant to treat metastatic estrogen-receptor positive, HER2-negative breast cancer after the cancer has grown despite hormone therapy plus a CDK4/6 inhibitor. It may help by finding a more effective treatment option after your first lines have stopped working.

Who can take part

  • You are female and age 18 or older.
  • Your breast cancer must be ER-positive (at least 1% by lab testing) and HER2-negative.
  • Your cancer must have grown after first-line hormone (endocrine) treatment plus a CDK4/6 inhibitor; you may have had letrozole before.
  • In metastatic disease, you should have had only one prior hormone (endocrine) therapy line, and no more than one prior chemotherapy line.
  • You must have measurable tumors on scans, be well enough to do daily activities (ECOG 0–1), and expect to live at least 12 weeks.
  • You must have test results that show good blood counts, liver and kidney function, and normal thyroid tests (or thyroid that can be controlled with treatment).
