This trial tests if lorlatinib, a targeted pill, can shrink or slow the growth of high-grade brain tumors that have a specific ROS or ALK genetic change. It is for children and young adults who have not yet had treatment for their tumor.

Who can take part
- You are between 1 and 21 years old.
- You have a newly diagnosed high-grade glioma (including DIPG) with an ALK or ROS-1 fusion.
- You have not received any chemotherapy or other cancer treatment before this study.
- You are able to take pills by mouth and do not have a condition that interferes with absorption.
- You have adequate organ function (bone marrow, kidney, liver, lungs, heart) as determined by tests.
- If you are female and of childbearing potential, you agree to use effective non-hormonal birth control during treatment and for 6 months after.
