This trial tests a targeted drug (entrectinib) in very young children (under 3) with a newly diagnosed brain tumor that has a specific genetic change (NTRK or ROS1 fusion). The drug works by blocking signals that help the tumor grow, and is given as the first treatment, before any chemotherapy or radiation.

Who can take part

  • Your child must be younger than 3 years old at the time of diagnosis.
  • The tumor must be in the upper part of the brain (supratentorial) and have a specific genetic change (NTRK1/2/3 or ROS1 fusion).
  • Your child cannot have had any prior chemotherapy or radiation therapy for this tumor.
  • Your child must be in good overall health, with adequate blood counts, kidney, and liver function.
  • Surgery (biopsy or partial removal) must have been done within the last 84 days, and there must be enough tumor tissue available for testing.
